Intended use


The Male Fertility (Sperm) Test Kit (Colloidal Gold) is a rapid chromatographic immunoassay for in vitro qualitative detection of acrosomal protein SP-10 found on sperms to estimate of sperm concentration in human semen above or below 15 million/ml. Sperm concentration can be used for assistant diagnosis and curative effect observation of male infertility, and provide guidance for reproductive planning for couples.



Test principle


This reagent uses immunochromatography technology and applies the double-antibody sandwich method to detect the specific protein SP10 in human semen. If the concentration of SP10 protein in semen samples is found to be higher than or equal to the limit of detection, the sperm SP10 protein in semen reacts with colloidal gold immunolabeling SP10 monoclonal antibody 1 to form a complex. Under chromatography, this complex moves forward along the nitrocellulose membrane, is captured by SP10 monoclonal antibody 2 pre coated in the test zone (T) of the nitrocellulose membrane, and ultimately forms a visible red reaction line, indicating a positive result. On the contrary, when the semen sample does not contain sperm SP10 protein or the concentration of sperm SP10 protein is below the limit of detection, no red reaction line appears in the test zone (T), indicating a negative result. Specimen requirements

                                                                                                                     

1. Wait at least 48 hours, but not more than seven days after your last ejaculation. This ensures that the volume and quality of sperm is at its peak and the test will then be an accurate determination of sperm concentration.

2. Obtain the semen sample by manual stimulation (masturbation) or other proper ways. Collect the sample in the Collection cup.

3. After collecting the sample, let the Collection cup stand upright on a flat surface.

4. The semen should be tested within 3 hours after collection.


Note:

【1】Care should be taken that collected semen is not contaminated by touch of hands or tissues or any other materials.

【2】Do not use any lubricants or lotions since this may interfere with the test result.



Test Procedure


Carefully read the reagent instruction before using the test kit and strictly operate according to the instruction to ensure reliable results. Bring test device, buffer and specimens were restored to room temperature 15-30℃(59-86℉).


1.Specimen Preparation


(1)Let semen stand for at least 20 minutes at room temperature before testing as semen is too thick to be tested immediately. Gently stir the semen sample with pipette in the cup about 10 times until it is well mixed.



(2)Transfer 0.1mL the semen sample into Buffer bottle with a Semen transfer device, avoiding any solid or sticky material within the semen. Avoid getting air bubbles in the tip. If this happens, push the semen back out completely and then draw semen into the tip again.


(3)Tighten the cap back onto the Buffer bottle and gently mix the contents thoroughly. This is best done by turning the bottle upside down at least five to ten times. If your semen is very thick or viscous, you should mix an additional ten times. Let the bottle containing the semen mixture stand for two (2) minutes before proceeding to the next step.




2.Testing


(1)Unscrew the cap on the buffer bottle, hold the sample dropper vertically,and add 2~3 full drops of the specimen without air bubbles into the sample well (S) on the testing device.


(2)Start the timer. Wait for the colored line(s) to appear. Read the results in 15 minutes.Do not interpret the result after 20 minutes.
